Endothelin-1 downregulates sperm phagocytosis by neutrophils in vitro: a physiological implication in bovine oviduct immunity OAK
MAREY, Mohamed Ali; YOUSEF, Mohamed Samy; LIU, Jinghui; MORITA, Kazuhiro; SASAKI, Motoki; HAYAKAWA, Hiroyuki; SHIMIZU, Takashi; ELSHAHAWY, Ibrahim I.; MIYAMOTO, Akio; 佐々木, 基樹; 清水, 隆; 宮本, 明夫.
The oviduct is an active contractile tube that provides the proper environment for sperm transport, capacitation and survival. Oviductal contractions are regulated by autocrine/paracrine secretion of several factors, such as prostaglandins (PGs) and endothelin-1 (EDN-1). We have previously shown that during the preovulatory stage, sperm are exposed to polymorphonuclear neutrophils (PMNs) in the bovine oviduct, and the bovine oviduct epithelial cells (BOECs) secrete molecules including PGE2 that suppress sperm phagocytosis by PMNs in vitro. In this study, we investigated the possible effects of EDN-1 on the phagocytic activity of PMNs toward sperm. The local concentrations of EDN-1 in oviduct fluid and BOEC culture medium ranged from 10–10 to 10–11 M as...

Etude des amibes marines et de leurs interactions avec les vibrios pathogènes d’huître ArchiMer
Robino, Etienne.
Free living amoebae inhabit aquatic environments and use phagocytosis of bacteria for their nutrition. According to the hypothesis of coincidental evolution of virulence, the cellular and molecular mechanisms of phagocytosis being preserved from amoebae to the immune cells of animals, the predation exerted by amoebae could favor the emergence of pathogenic bacteria resistant to phagocytosis. Since 2008, Crassostrea gigas oysters have suffered from over-mortality in France. This poly-microbial disease involves the Herpes OsHV-1 μvar virus which causes an immunosuppression of oysters that are then colonized by various opportunistic pathogenic bacteria including vibrios inducing the death of the animal. V. tasmaniensis LGP32 is a facultative intracellular...

Etude des effets de polluants sur les paramètres hémocytaires de l'huître creuse, Crassostrea gigas - Interactions entre environnement, mécanismes de défense et maladies infectieuses ArchiMer
Gagnaire, Beatrice.
Shellfish industry is mostly realized in estuary zones, which are subjected to pollutions due to anthropic activities. The harmful effects of pollutants on animals inhabiting these estuarine zones are poorly known. Among these animals, Pacific oyster, Crassostrea gigas, may represent a model because they are sedentary and they filter water intensively. Among all physiological functions possibly disturbed by pollutants, defence mechanisms are poorly studied in bivalves. Moreover, animals presenting impaired defence mechanisms may be more sensitive to infectious diseases. In this context, effects of pollutants on hemocyte functions and on sensitiveness to diseases were tested in C. gigas. After adjusting the protocols for the monitoring of hemocyte...

Evaluation of possible failure of the mononuclear phagocyte system after total splenectomy in rats BABT
Marques,Ruy Garcia; Petroianu,Andy; Oliveira,Márcia Betânia Nunes de; Bernardo-Filho,Mário; Portela,Margareth Crisóstomo.
Young and adult Wistar rats were submitted to total splenectomy and compared to animals not submitted to any surgical manipulation in order to evaluate the phagocytic function of spleen. The animals were infected with Escherichia coli labeled with technetium-99m and killed 20 minutes later. Liver, lung, spleen and a blood clot sample were taken. No significant differences were found in the percentage of bacterial radioactivity uptake in mononuclear phagocyte system (MPS) organs in young and adult splenectomized rats. However, phagocytosis index by macrophages of MPS organs was smaller in splenectomized animals than in control group. Splenectomized rats were associated with a higher blood bacterial radioactivity uptake than animals of the control group...

Fine cytochemical localization of lysosomal organelles in phagocytic cells of bivalve molluscs. ArchiMer
Auffret, Mathieu; Diouris, M; Herry, A.
Bivalve molluscs possess several types of hemolymph cells (hemocytes), involved in internal defense and other physiological functions. Studies in pathological and experimental conditions have shown their micro- and macrophagic ability. However, the mechanisms of intracellular digestion are incompletely known in these organisms. The lysosomal apparatus of hemocytes was therefore investigated in several species by hydrolase localization with the electron microscope. The cytoplasmic granules of granulocytes displayed a strong acid beta -glycerophosphatase activity. Previous observations had shown that these granules may contain phagocytized material. This indicates that intracellular digestion is achieved in these organelles. Acid beta -glycerophosphatase...

FISH IMMUNE SYSTEM Gayana
Olabuenaga,Susana E..
The defence against infections is the essential function of the immune system of all Vertebrates. Fish present an efficient and developed immunologic response and in the case of Teleosts, with some similarities to the higher vertebrates. It can be divided into two types: innate or non specific and adquired or specific, both with a humoral and cellular compartments. Their relative importance can change according to the age of fish and is always under the influence of different factors. The most relevant components are mentioned here. There are two factors in fishes which strongly influence the immune response and antibody production, seasonal changes and temperature. The main lymphoid organs are described here: the thymus, the kidney and the spleen, as well...

Fluorescent membrane markers elucidate the association of Borrelia burgdorferi with tick cell lines BJMBR
Teixeira,R.C.; Baêta,B.A.; Ferreira,J.S.; Medeiros,R.C.; Maya-Monteiro,C.M.; Lara,F.A.; Bell-Sakyi,L.; Fonseca,A.H..
This study aimed to describe the association of Borrelia burgdorferi s.s. with ixodid tick cell lines by flow cytometry and fluorescence and confocal microscopy. Spirochetes were stained with a fluorescent membrane marker (PKH67 or PKH26), inoculated into 8 different tick cell lines and incubated at 30°C for 24 h. PKH efficiently stained B. burgdorferi without affecting bacterial viability or motility. Among the tick cell lines tested, the Rhipicephalus appendiculatus cell line RA243 achieved the highest percentage of association/internalization, with both high (90%) and low (10%) concentrations of BSK-H medium in tick cell culture medium. Treatment with cytochalasin D dramatically reduced the average percentage of cells with internalized spirochetes,...

Hemócitos fagocitários em larvas de Diatraea saccharalis (Fabricius) (Lepidoptera, Pyralidae) Zoologia
Falleiros,Angela M.F; Gregório,Elisa A.
The ultrastructural observation of the hemocytes of D. saccharalis larvae showed different hemocyte tvpes in various stage of interaction with microorganisms, which were interpreted as bacteria. The microorganisms were observed either free in the hemolymph or into the insect hemocytes. The bacteria were detected mostly into lhe granulocytes and plasmatocytes: the spherule cells and the oenocytoids also presented intracytoplasmic microorganisms, but less frequently. The fagocytic capacity of the different hemocyte tvpes is discussed.

Human monocytes tolerant to LPS retain the ability to phagocytose bacteria and generate reactive oxygen species BJMBR
Fernandes,M.L.; Mendes,M.E.; Brunialti,M.K.C.; Salomão,R..
Tolerance to lipopolysaccharide (LPS) occurs when animals or cells exposed to LPS become hyporesponsive to a subsequent challenge with LPS. This mechanism is believed to be involved in the down-regulation of cellular responses observed in septic patients. The aim of this investigation was to evaluate LPS-induced monocyte tolerance of healthy volunteers using whole blood. The detection of intracellular IL-6, bacterial phagocytosis and reactive oxygen species (ROS) was determined by flow cytometry, using anti-IL-6-PE, heat-killed Staphylococcus aureus stained with propidium iodide and 2',7'-dichlorofluorescein diacetate, respectively. Monocytes were gated in whole blood by combining FSC and SSC parameters and CD14-positive staining. The exposure to...

Immune priming and portal of entry effectors improve response to vibrio infection in a resistant population of the European abalone ArchiMer
Dubief, Bruno; Nunes, Flavia; Basuyaux, Olivier; Paillard, Christine.
Since 1997, populations of the European abalone Haliotis tuberculata suffer mass mortalities attributed to the bacterium Vibrio harveyi. These mortalities occur at the spawning season, when the abalone immune system is depressed, and when temperatures exceed 17 °C, leading to favorable conditions for V. harveyi proliferation. In order to identify mechanisms of disease resistance, experimental successive infections were carried out on two geographically distinct populations: one that has suffered recurrent mortalities (Saint-Malo) and one that has not been impacted by the disease (Molène). Furthermore, abalone surviving these two successive bacterial challenges and uninfected abalone were used for several post-infection analyses. The Saint-Malo population...

Immune response of the Antarctic sea urchin Sterechinus neumayeri: cellular, molecular and physiological approach ArchiMer
Gonzalez-aravena, Marcelo; Perez-troncoso, Carolina; Urtubia, Rocio; Branco, Paola; Machado Cunha Da Silva, Jose Roberto; Mercado, Luis; De Lorgeril, Julien; Bethke, Jorn; Paschke, Kurt.
In the Antarctic marine environment, the water temperature is usually between 2 and - 1.9 degrees C. Consequently, some Antarctic species have lost the capacity to adapt to sudden changes in temperature. The study of the immune response in Antarctic sea urchin (Sterechinus neumayeri) could help us understand the future impacts of global warming on endemic animals in the Antarctic Peninsula. In this study, the Antarctic sea urchins were challenged with lipopolysaccharides and Vibrio alginolitycus. The cellular response was evaluated by the number of coelomocytes and phagocytosis. A significant increase was observed in red sphere cells and total coelomocytes in animals exposed to LPS. A significant rise in phagocytosis in animals stimulated by LPS was also...

Impact of food availability on energy storage and defense related hemocyte parameters of the Pacific oyster Crassostrea gigas during an experimental reproductive cycle ArchiMer
Delaporte, Maryse; Soudant, P; Lambert, C; Moal, Jeanne; Pouvreau, Stephane; Samain, Jean-francois.
The aim of this study was to test the effect of food quantity on energy storage and defense capacities of oysters during a reproductive cycle. One-year-old Crassostrea gigas oysters were fed two different dietary rations (4% and 12% of oyster dry weight in algal dry weight per day) in controlled experimental conditions over an annual cycle. Oyster dry weights, carbohydrate and lipid contents, energetic adenylate charge, and hemocyte parameters of oysters were significantly affected by reproductive processes related to seasonal temperature variation and, to a lesser extent, by the dietary rations. Energy parameters decreased during gametogenesis as gonads developed then increased during the gonad resorption phase. The additional energy provided to oysters...

Increased leishmanicidal activity of alveolar macrophages from mature horses with mild equine asthma Arq. Bras. Med. Vet. Zootec.
Lessa,D.A.B.; Alencar,N.X.; Torres Filho,R.A.; Costa,M.F.M.; Fernandes,W.R.; Saraiva,E.M..
ABSTRACT Alveolar macrophages (AMs) are an essential part of defense mechanisms within the lungs and their phagocytic activity is important for organ homeostasis. The phagocytic ability of AMs obtained from bronchoalveolar lavage from 17 mature mixed-breed pleasure horses (8 healthy and 9 diagnosed with mild equine asthma) was studied through assays with Leishmania (Viannia) braziliensis promastigotes, which enabled the calculation of a phagocytic index (PI) and a survival index (SI). Results indicate that phagocytic activity of AMs in asthma affected horses is similar to healthy horses, while leishmanicidal activity is significantly increased in horses with asthma.

Influence of acute renal failure on the mononuclear phagocytic system BJMBR
Sousa,V.R.A.; Sousa,A.A.; Petroianu,A.; Simal,C.J.R.; Barbosa,A.J.A..
Several studies show the ability of macrophages to remove particles injected into the bloodstream. This function seems to be increased in the presence of acute renal failure. The objective of the present study was to assess the phagocytic function of the main organs (spleen, liver and lung) of the mononuclear phagocytic system in renal and postrenal failures. Fifteen rats (250-350 g) were divided into three groups (N = 5): group I - control; group II - ligature of both ureters, and group III - bilateral nephrectomy. On the third postoperative day, all animals received an iv injection of 1 ml/kg 99mTc sulfur colloid. Blood samples were collected for the assessment of plasma urea, creatinine, sodium, and potassium concentrations and arterial gasometry....

Influence of orchiectomy of seven month old bulls on bronchoalveolar immune function Arq. Bras. Med. Vet. Zootec.
Bertagnon,H.G.; Batista,C.F.; Santos,B.P. dos; Lima,M.G.B.; Bellinazzi,J.B.; Libera,A.M.M.P. Della.
ABSTRACT The present study evaluated the impact that orchiectomy, a routine but painful intervention in bovine husbandry, can cause on pulmonary immunity. To identify whether orchiectomy can impair lung defense, analyses of serum cortisol concentration and of alveolar macrophage and their function (phagocytosis and respiratory burst) were evaluated. Sixteen Holstein bulls (7 mo old, 250±50kg of body weight BW) were divided in two homogeneous groups - the castrated group and the sham group - and the sample were collected on Days -7, 1 and 7 relative to the day of the procedure. Serum cortisol concentration statistically increased on Days 1 and 7 (D-7: 4,97±1,28ng/ml; D1: 6,35 ±1,10ng/ml; D7: 8,28±1,94ng/ml) after castration and these variables seem to...

MFG-E8, a clearance glycoprotein of apoptotic cells, as a new marker of disease severity in chronic obstructive pulmonary disease BJMBR
Zhang,S.; Xie,J.G.; Su,B.T.; Li,J.L.; Hu,N.; Chen,J.; Luo,G.W.; Cui,T.P..
Milk fat globule epidermal growth factor 8 (MFG-E8) is an opsonin involved in the phagocytosis of apoptotic cells. In patients with chronic obstructive pulmonary disease (COPD), apoptotic cell clearance is defective. However, whether aberrant MFG-E8 expression is involved in this defect is unknown. In this study, we examined the expression of MFG-E8 in COPD patients. MFG-E8, interleukin (IL)-1β and transforming growth factor (TGF)-β levels were measured in the plasma of 96 COPD patients (93 males, 3 females; age range: 62.12±10.39) and 87 age-matched healthy controls (85 males, 2 females; age range: 64.81±10.11 years) using an enzyme-linked immunosorbent assay. Modulation des paramètres hémocytaires par la nutrition de l'huitre creuse Crassostrea gigas. Implication dans les mortalités estivales ArchiMer
Delaporte, Maryse.
In the context of Pacific oyster C. gigas summer mortality, the present study demonstrated that the reproductive effort reduced not only the energetic parameters (carbohydrates and CEA) but also the immune parameters (concentration, phagocytosis and adhesive capacity of hemocytes). Increasing food supply from 4% to 12% (of oyster dry weight) did not counteract this phenomena as most of the additional energy was dedicated to the gametogenesis processes. However, after the gametogenesis, the 12% diet clearly favored energy storage restoration and hemocyte production. Hemocyte parameters were also modulated by the food quality. Indeed, hemocyte concentration tended to increase 20:5n-3 and/or 20:4n-6 were supplied by micro-algae or artificial diets. When these...

Modulation of peritoneal macrophage activity by the saturation state of the fatty acid moiety of phosphatidylcholine BJMBR
Grando,F.C.C.; Felício,C.A.; Twardowschy,A.; Paula,F.M.; Batista,V.G.; Fernandes,L.C.; Curi,R.; Nishiyama,A..
To determine the effects of saturated and unsaturated fatty acids in phosphatidylcholine (PC) on macrophage activity, peritoneal lavage cells were cultured in the presence of phosphatidylcholine rich in saturated or unsaturated fatty acids (sat PC and unsat PC, respectively), both used at concentrations of 32 and 64 µM. The treatment of peritoneal macrophages with 64 µM unsat PC increased the production of hydrogen peroxide by 48.3% compared to control (148.3 ± 16.3 vs 100.0 ± 1.8%, N = 15), and both doses of unsat PC increased adhesion capacity by nearly 50%. Moreover, 64 µM unsat PC decreased neutral red uptake by lysosomes by 32.5% compared to the untreated group (67.5 ± 6.8 vs 100.0 ± 5.5%, N = 15), while both 32 and 64 µM unsat PC decreased the...
